MBX-4132, which interferes with protein synthesis by blocking trans translation, has several characteristics that could make it a match for N. gonorrhoeae. In preclinical experiments, the team showed that the bug is slow to develop resistance to the drug – an unusual feature for N. gonorrhoeae, which is famous for the rapid development of resistance. While it is not clear what makes MBX-4132 such a challenge for N. gonorrhoeae, the bug has four genetic copies of the ribosome, and Aron speculated that possibly, N. gonorrhoeae needs to develop resistance mutations in each of the copies to become resistant.Another promising feature of the drug is that in animal studies, it shows significant efficacy after a single dose. In one animal study, a single oral dose of MBX-4132 cured 70 percent of mice who had multidrug-resistant gonorrhea.
